Significant advances have been made in understanding the mechanistic underpinnings of energy homeostasis and obesity in laboratory animal models. Specific genes that are essential for body weight regulation have been identified, and mutations to some of these genes constitute the causative lesion in obese human subjects. Despite the advances that have been made in studying monogenic models of obesity, it is clear that most human obesity is polygenic and represents the interaction between multiple genes and environment, of which diet is a major component. Accordingly, there is a pressing need to investigate polygenic rodent models of obesity, with diet-induced obesity DIO ; 1 models coming under increasing scrutiny. In another placebo-controlled trial, Alzheimer's disease patients treated with Reminyl died at a significantly higher rate than those who received placebos. Since Aricept, Reminyl, and similar drugs slow the heart and constrict the respiratory passages, the findings of these trials are unlikely to be flukes.20 268.
